Longest single-fluorescent-molecule tracking ever reported in living cells with SeTau-647 - Nature Chem. Biol. (2018)

The photobleaching rates of 13 fluorescent dyes, 4 of them conjugated to trolox, were investigated for single-molecule measurements at 37oC in living cells. Among these SeTau-647 (K9-4149 and K9-4148), a squaraine rotaxane dye, exhibited the best photobleaching performance in this study with an exponential lifetime of 105.8 sec. As a membrane-impermeable dye it allowed to observe up to 12,000 frames, which is the longest single-fluorescent-molecule tracking ever reported. See paper in Nat. Chem.
